چکیده

The development of immunosuppressive agents with less toxicity and more specificity has led researchers to explore the role of interleukin-2 (IL-2) in the immunosuppression cascade. The IL-2 receptor is composed of at least 3 subunits. One subunit, the a chain, named the Tac antigen, is predominantly expressed on activated T cells. Interleukin-2 facilitates the proliferation of alloantigen-activated T-cell clones by interacting with the IL-2 receptor, a critical step in the process of allograft rejection. For this reason, monoclonal antibody development was directed at this portion of the receptor and was called anti-Tac. Agents such as cyclosporine and tacrolimus target different sites along the immunosuppression cascade. It is thought that the IL-2 receptorblocking monoclonal antibody will produce a synergistic effect when given in addition to current 3-drug regimens (i.e., cyclosporine or tacrolimus plus steroids plus azathioprine or mycophenolate mofetil) (1–3).

Production and Evaluation of Specific Single-Chain Antibodies against CTLA-4 for Cancer-Targeted Therapy

Background:  Cytotoxic T lymphocyte–associated antigen 4 (CTLA-4) molecules are expressed on T-cells and inhibit their function by inhibiting activation of subsequent T-cell molecular pathways. Blocking of CTLA-4 inhibits the growth of malignant tumor cells. Anti-CTLA-4 monoclonal antibodies activate the immune system against cancer. Due to several advantages of single-chain antibodi...

Blocking the receptor for interleukin 10 protects mice from lethal listeriosis.

High doses of Listeria monocytogenes overcome the ability of a normal mouse to control the infection, due to massive bacterial replication. Treatment with an anti-interleukin 10 (IL-10) receptor monoclonal antibody prevented the fatal course of infection with high doses of bacteria. This work shows that blocking the receptor for IL-10 may have useful therapeutic applications.

Aspergillosis in patients treated with monoclonal antibodies.

Invasive aspergillosis is a disease typically related with prolonged neutropenia or the use of corticosteroids. However, the increased use of new therapeutic modalities such as biologic agents that act by blocking specific immune pathways have put more patients at risk for invasive aspergillosis. Most cases of aspergillosis in patients taking monoclonal antibodies have been associated with the ...

The impact of biological agents interfering with receptor/ligand binding in the immune system.

We herein discuss the impact of biological agents based on the ability of monoclonal antibodies to target specific molecules. This approach has given to clinical immunologists a spectrum of drugs able to manipulate the immune system. In the first session, we discuss drugs targeting T-cell function by: (1) targeting CD28 mediated costimulation (Abatacept and Belatacept); (2) interfering with int...

The measurement technologies of thyrotropin receptor antibodies from the past to the present

Thyroid Stimulating Hormone Receptor (TSH-R) autoantibodies are the main cause of Graveschr('39') disease and its external thyroid manifestations such as ophtalmopathy and dermatopathy. These antibodies are functionally different and are commonly called TSH receptor antibodies (TRAbs). In fact, TRAbs are a set of autoantibodies including TSHR-stimulating antibodies (TSAbs), TSHR- blocking antib...
